Scientists have identified a new immune structure in the bone marrow of the skull that acts like a security checkpoint, protecting the brain against tumors and diseases. The discovery, published in the journal Nature, could open a new era in the treatment of many neurological conditions, especially deadly brain cancers. Researchers at Washington University School of Medicine found that the skull marrow is not just a protective bone framework but hosts critical centers that manage brain-specific immune responses.
By tracking micro-channels extending from the brain to the skull with fluorescent proteins, the team encountered structures resembling lymph nodes where immune cells are trained. These structures train and multiply defense cells directly inside the skull, allowing the body to mount a frontline fight against dangers without waiting for reinforcements from distant parts of the body. The power of this newly discovered structure was tested on mice carrying glioma, a deadly form of brain tumor.
When the immune center was suppressed with a special gel applied under the scalp, tumors spread much faster. Conversely, mice treated with a gel containing immune-boosting proteins saw their skull defense cells fight the tumor far more effectively, with a marked increase in survival rates. While currently proven in mice, there are strong indications that these special cells also exist in the human skull.
Researchers emphasize that if confirmed in humans, this discovery could transform the medical world. A simple injection under the scalp targeting the immune center directly could offer a far less risky treatment option compared to open brain surgery. This approach is expected to open new doors not only for brain tumors but also for immune-related brain diseases such as Alzheimer's, Parkinson's, and long COVID.
